General description

Tissue inhibitor of metalloproteinases 4 (UniProt: Q99727; also known as Metalloproteinase inhibitor 4, TIMP-4) is encoded by the TIMP4 gene (Gene ID: 7079) in human. The tissue inhibitors of metalloproteinases (TIMPs) represent a family of ubiquitous proteins which are natural inhibitors of the matrix metalloproteinases (MMPs). There are four known members of the TIMP family that are designated as TIMP-1, 2, 3, and 4. TIMP-4 is synthesized with a signal peptide (aa 1-29), which is cleaved off in the mature form. It has two MMP-binding regions (aa 30-33 and 99-100). TIMP-4 expressed is observed in heart, brain, pancreas, kidney, colon, testes, and adipose tissue. However, elevated levels of TIMP-4 have been reported in various cancers tissues, including breast, ovary, prostate, colon and kidney and are associated with poor survival prognosis. Hence, it can serve as a useful marker for disease progression. TIMP-4 is shown to complex MMPs and irreversibly inactivates them by binding to their catalytic zinc cofactor. This clone (9:4 7) is specific for human TIMP-4 and does not exhibit any cross-reactivity to other TIMPs. (Ref.: Gomez, DE., et al. (1997). Eur. J. Cell.
