General description

Autophagy related 7 (ATG7) is encoded by the gene mapped to human chromosome 3p25.3. The encoded protein has ubiquitous expression.
Monoclonal Anti-Atg7 (mouse IgG1 isotype) is derived from the hybridoma ATG7-13 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with a synthetic peptide corresponding to a sequence located near the N-terminus of human Atg7.

Immunogen

synthetic peptide corresponding to a sequence located near the N-terminus of human Atg7, conjugated to KLH. The corresponding sequence is identical in mouse and rat Atg7.

Application

Monoclonal Anti-Atg7 antibody produced in mouse has been used in:

Biochem/physiol Actions

Atg7 can activate all three Atg8 mammalian homologues, (Golgi-associated ATPase enhancer of 16 kDa (GATE-16), GABA receptor-associated protein (GABARAP) and microtubule-associated protein light chain 3 (MAP-LC3). It forms a homodimer through the C-terminal region that is important for enzyme substrate interaction and E1-E2 complex formation. Atg7 was found to be essential for amino acid supply in neonates and starvation− induced bulk degradation of proteins and organelles in mice. Loss of Atg7 results in the accumulation of abnormal organelles and ubiquitin positive aggregates and leads to neurodegeneration.
Autophagy related 7 (ATG7) plays a vital role in the regulation of adult hematopoietic stem cells (HSCs). ATG7 expression is elevated in eosinophilic esophagitis (EoE). Hence, this can be used as a potent tissue biomarker for diagnosis of EoE inflammation in pediatric patients. The encoded protein plays an essential role in autophagy vesicle elongation. In addition, it is also implicated in two vital ubiquitin-like reactions such as LC3 lipidation and Atg5/12 conjugation. Down regulated expression of Atg7 increases the sensitivity to chemotherapy and disables stroma-mediated chemoresistance in acute myeloid leukemia (AML).

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
