General description

Media are supplemented with a defined mixture of nutrients, growth factors, and hormones instead of serum. A 1:1 mixture of Dulbecco′s Modified Eagle′s medium (DME) and Ham′s F-12 nutrient mixture is commonly used for these defined studies. To compensate for the loss of buffering capacity caused by eliminating serum, a final concentration of 15 mM HEPES buffer can be added to the formulations. It allows the investigator to optimize the buffering system.

Application

Dulbecco′s Modified Eagle′s Medium/Nutrient Mixture F-12 Ham has been used:
  • to maintain human lung and colorectal cancer cell lines for the biological evaluation of the secondary metabolites
  • to culture human kidney-2 (HK-2) cells and as a component of the assay medium for lysophosphatidylcholine (LPC) treatment
  • to culture mouse squamous cell carcinoma (SCCVII) tumor cells
