General description

Anti-Caspase 8 antibody, Mouse monoclonal (mouse IgM isotype) is derived from the CAS8 hybridoma produced by the fusion of mouse myeloma cells and splenocytes from a BALB/c mouse immunized with a recombinant human caspase 8. Caspase 8 gene is mapped to human chromosome 2q33.1 Caspases (Cysteine-requiring aspartate proteases) are a family of proteases. The caspase 8 is also known as MORT1-associated ced-3 homolog (MACH), FLICE (FADD-homologous ICE/ced-3-like protease) and Mch5. Eight isoforms of caspase 8 (caspase 8/a-h), including FLICE (CAP4/MACHα1), MACHα2 and MACHα3, MACHβ and Mch5 have been described at the mRNA level. Caspase 8 contains two amino-terminal tandem repeats within the prodomain, which are homologous to the death effector domain (DED) in the adaptor molecule Fas-associated death domain protein (FADD). It belongs to the tumor necrosis factor receptor family.

Biochem/physiol Actions

Caspase 8 (CASP8) undergoes a cleavage upon activation (leading to p26/p24/p18/p10 subunits, for isoforms 8/a and 8/b). Caspase-8 and caspase-10 are involved in death receptor-mediated apoptosis. CASP8 is a crucial defensive barrier against malignant proliferation and tumorigenesis by triggering the extrinsic apoptosis. Polymorphisms in the CASP8 gene is also an indicative biomarker for lung and breast cancers. Caspase 8 is also implicated in invasive colon cancer. Caspase 8 has been shown to process a variety of proapoptotic caspases, but it did not activate members of the ICE subfamily, except of ERICE (caspase 13). Monoclonal antibodies reacting specifically with caspase 8 are a useful tool for the study of the protease network involved in development and regulation, governing the life and death of cells and tissues.
Caspases (Cysteine-requiring Aspartate protease) are a family of proteases. Caspase-8 and -10 are involved in death receptor mediated apoptosis.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2–8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in "frost-free" freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
