General description

Nucleobindin-2 (UniProt Q9JI85; also known as DNA-binding protein NEFA, Prepronesfatin) is encoded by the Nucb2 (also known as Nefa) gene (Gene ID 59295) in rat species. Nucleobindin-2 is initially translated as a 420-a.a. precursor protein. Cleavage of signal peptide (aa1-24) yields the 396-a.a. mature nucleobindin-2 with very high sequence homology among human, mouse, and rat species (more than 85%). Post-translational processing by prohormone convertases (PC 1/3 and PC 2) cleaves nucleobindin-2 into nesfatin-1, nesfatin-2, and nesfatin-3 (aa1–82, aa85–163, and aa166–396, respectively, numbering based on mature sequence). Nesfatin-1 is a well recognized circulating anorexigenic peptide, whose production increases post-prandially. Nucleobindin-2 contains two calcium-binding EF-hand domains separated by an acidic amino acid-rich region, and a leucine zipper. Nucleobindin-2 is known to be expressed in key hypothalamic nuclei and plays a role in energy homeostasis. Nucleobindin-2 is also expressed in various human peripheral tissues, including the stomach, pancreas, reproductive organs, and adipose tissues, suggesting roles in various metabolic functions. Nucleobindin-2 expression level is reported to be elevated in clear cell renal cell carcinoma (ccRCC), breast cancer, and prostate cancer (PCa) tissues.

Specificity

Clone 3B3.1 reacts with pro-nucleobindin-2, mature nucleobindin-2, and nesfatin-1, but not nesfatin-2 or nesfatin-3.

Epitope: Nesfatin-1
GST-tagged recombinant rat Nestafin-1.

Application

Immunohistochemistry Analysis: A 1:50-250 dilution from a representative lot detected nucleobindin-2/nestafin-1 immunoreactivity in human colon cancer tissue, as well as in normal testis, tonsil, and pancreas tissue sections.

Quality

Evaluated by Western Blotting in mouse liver tissue lysate.

Western Blotting Analysis: 0.5 µg/mL of this antibody detected nucleobindin-2 in 10 µg of mouse liver tissue lysate.
