General description

Syntaphilin (SNPH) is a single-pass membrane protein that was initially identified in a yeast two-hybrid screen with the carboxy terminal region of Syntaxin-1 as bait. Syntaxin-1 is a key component of the synaptic vesicle docking machinery that forms the SNARE complex with synaptobrevin and SNAP-25. Syntaphilin competes with SNAP-25 for binding to syntaxin-1 and inhibits the formation of the SNARE complex, thereby potentially regulating synaptic vesicle exocytosis. Syntaphilin also binds dynamin-1 and inhibits dynamin-dependent endocytosis. Mice lacking syntaphilin show an increased level of mitochondrial motility and a reduced density of axonal mitochondria. This correlates with an enhanced short-term facilitation and significant impairments in motor ability, suggesting syntaphilin plays a major role in presynaptic function.

Immunogen

Epitope: center region
KLH-conjugated synthetic peptide corresponding to the center region of human Syntaphilin.

Application

Anti-Syntaphilin Antibody is an antibody against Syntaphilin for use in western blotting, IHC & immunofluorescence.
Immunohistochemistry Analysis: 5 μg/mL from a representative lot detected Syntaphilin in human brain tissue.

Immunofluorescence Analysis: 20 μg/mL from a representative lot detected Syntaphilin in human brain cells.

Quality

Evaluated by Western Blotting in rat brain tissue lysate.

Western Blotting Analysis: 2 μg/mL of this antibody detected Syntaphilin in rat brain tissue lysate.

Target description

~86 kDa observed. Uncharacterized bands may be observed in some lysate(s). Despite its predicted molecular weight, Syntaphilin usually migrates at higher molecular weight in SDS-PAGE.

Physical form

Antigen Affinity Purified
Purified rabbit polyclonal in buffer containing PBS with up to 0.1% sodium azide.

Storage and Stability

Stable for 1 year at 2-8°C from date of receipt.
