Clone 1M23 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects Homeobox protein SIX1 (SIX1). It targets an epitope within 25 amino acids from the C-terminal half of the protein.

Immunogène

KLH-conjugated linear peptide corresponding to 25 amino acids from the C-terminal half of human Homeobox protein SIX1.

Application

Quality Control Testing

Evaluated by Western Blotting in L6 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Homeobox protein SIX1 (SIX1) in L6 cell lysate.

Tested Applications

Affinity Binding Assay Analysis: The binding affinity of this antibody to SIX1 peptide was at least ten thousand-fold (10000X) higher than that obtained with the non-specific control peptide.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected SIX1 in human cervix tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected SIX1 in C2C12 cells.

Description de la cible

Homeobox protein SIX1 (Uniprot: Q15475; also known as Sine oculis homeobox homolog 1, SIX1) is a transcription factor that is encoded by the gene SIX1 (Gene ID: 6495) in human. SIX1 belongs to a family of evolutionarily conserved transcription factors that are found in lower and higher organisms. A total of six family members (SIX1-6) have been identified in vertebrates that contain a homeobox nucleic acid recognition domain (HD) with a DNA-binding helix-turn-helix motif and an adjacent SIX domain, which may be involved in regulating protein-protein interactions. It can function both as a transcriptional repressor or activator depending on the context. This protein plays a crucial role in the regulation of cell proliferation, apoptosis, and embryonic development. It acts by regulating the expression of target genes involved in organogenesis and morphogenesis. SIX1 is primarily known for its role in the development of multiple organs, such as the kidney, ear, and skeletal muscles. SIX1 mediates apoptosis, autophagy, and regulates resistance to the drug in hepatocellular carcinoma. It is highly expressed in colorectal cancer and promotes tumor progression. Abnormal expression of SIX1 is also associated with other cancer subtypes, including Hodgkin lymphoma, breast, and ovarian cancer. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
