Clone 1G22 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Peroxiredoxin 1. It targets an epitope within 17 amino acids from the internal region.

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Peroxiredoxin 1 in HeLa cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Peroxiredoxin 1 in lysates from HepG2 and Raw264.7 cells and Rat heart tissue.

Affinity Binding Assay: A representative lot of this antibody bound Peroxiredoxin 1 peptide with a KD of 9.3 x 10-7 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Peroxiredoxin 1 in human thyroid tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Peroxiredoxin-1 (UniProt: Q06830; also known as EC:1.11.1.15, Natural killer cell-enhancing factor A, NKEF-A, Proliferation-associated gene protein, PAG, Thioredoxin peroxidase 2, Thioredoxin-dependent peroxide reductase 2) is encoded by the PRDX1 (also known as PAGA, PAGB, TDPX2) gene (Gene ID: 5052 ) in human. Peroxiredoxins belong to a family of non-seleno peroxidases. Thus far six different mammalian isoforms have been reported that catalyze peroxide reduction of hydrogen peroxide, organic hydroperoxides and peroxynitrite. They all share a conserved Cys51 residue in the N-terminal region. Peroxiredoxin 1-4 also contain an additional conserved Cys residue in the C-terminal region, Cys172. The N-terminal Cys51 is oxidized by hydrogen peroxide to form cysteine-sulfenic acid (Cys51-SOH) and reacts with Cys172-SH of the other subunit to produce an intermolecular disulfide, which can only be reduced by thioredoxin and not by glutathione or glutaredoxin. Peroxiredoxin-1, a thiol-peroxidase, is constitutively expressed in most human cells and is induced to higher levels upon serum stimulation of transformed and untransformed cells. It is involved in redox regulation of the cell and plays an important role in overcoming oxidative stress. Peroxiredoxin-1 is also involved in eliminating peroxides generated during drug metabolism. It is phosphorylated on Thr90 during the M-phase of cell cycle, which leads to over 80% reduction in its enzymatic activity. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
