Description générale

Nucleobindin 2 (NUCB2), also known as DNA binding/EF-hand/acidic protein (NEFA), p54, is encoded by the gene mapped to human chromosome 11p15. It belongs to the nucleobindin family of EF-hand Ca2+ - binding proteins. NUCB2 contains multiple functional domains, including a Leu/Ile rich region, a putative nuclear localization signal, a DNA binding domain, two Ca2+ -binding EF-hand motifs and a leucine zipper region. NUCB2 is widely distributed and is localized to the Golgi apparatus. NUCB2 has been shown to interact with both necdin and ARTS-1, through the EF-hand motif.

Actions biochimiques/physiologiques

Nucleobindin 2 (NUCB2) has been implicated in various functions, including Ca2+ homeostasis, hypothalamic regulation of feeding and tumor necrosis factor (TNF)-receptor-1 (TNFR1)shedding. The NUCB2-ARTS-1 complex is essential for the proteolytic cleavage and release of TNFR1 to the extracellular compartment. NUCB2 has been identified as a novel satiety molecule in the hypothalamus associated with melanocortin signaling in the central nervous system. NUCB2/nesfatin-1 has been shown to induce suppression of food-intake in rats.

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.

Stockage et stabilité

Store at –20 °C. For continuous use, the product may be stored at 2–8 °C for up to one month. For extended storage, freeze in working aliquots at –20 °C. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
