General description

The Adenosine 5′-triphosphate (ATP) Bioluminescent Assay Kit may be employed for the quantitative bioluminescent determination of ATP in samples. ATP is consumed and light is emitted when luciferase catalyzes the oxidation of D-luciferin. When ATP is the limiting reagent, the light emitted is proportional to the ATP present in the sample.

Application

Adenosine 5′-triphosphate (ATP) Bioluminescent Assay Kit has also been used in the quantification of ATP in 3D matrixes of human neurons,[1] hepatic cells with ischemia,[2] various bacterial cultures[3] and lysosomes.[4]

  1. How many times can one do with Product FLAAS, ATP Bioluminescent Assay Kit (one vial of FLAAS) when standard range between 20uM~0.002uM?

    1. The number of times that one can use FLAAS ATP Assay Kit is dependent upon the sample. Frequently, serial dilutions of the sample, as well as the kit reagents, are required. If no dilutions were required the ATP Assay MIx is reconstituted to 5 mL and added to the sample at 0.1 mL. The dilution factor will need to be determined by the end-user. Please note, FLAAS is stable for at least 24 hours at 2–8 °C, or over two weeks if stored frozen at –20 °C. Working concentrations of the serially diluted standard are stable for up to 8 hours when stored in ice.

  2. How does the kit perform on samples with pH 7.3-7.4 (physiological range)? It recommends adjustment of sample pH to 7.8 which is not always possible.

    1. It is important to use correct pH-value. The luciferase reaction has its pH-optimum in the range of 7.75. Samples having extreme high pH-values or samples of a different pH containing high buffer concentrations may influence the light reaction in an unpredictable way. To avoid issues, correct the pH to a value between 7.6 and 8.0 or dilute samples in Tricine Buffer of the appropriate pH.

  3. Can Product FLAA, ATP Bioluminescent Assay Kit assay mix be used on cells and tissues?

    1. This reagent measures only soluble ATP. Cells must be washed and then lysed to obtain a soluble ATP extract. Tissues must be homogenized and clarified to provide an extract. The supernatant solution can then be used for assay.

    1. With FLAAM diluted 1:5, one can easily measure 1 pmol ATP in 100 μl (10 nM) with 50 μl of the reagent.

    1. When one reconstitutes the vial to 5 ml, as directed, one can perform 50 assays using 100 ul per test. If one dilutes 1:5 one can get 250 tests per kit.
