Application
La carboxypeptidase B a été utilisée dans une étude visant à mettre au point un test de grossesse pour les ours polaires en captivité ou sauvages. Elle a également été utilisée dans une étude qui a identifié de nouveaux biomarqueurs potentiels de la pancréatite aiguë.
L′enzyme proposée par Sigma a été utilisée pour développer un essai HTRF (homogeneous time-resolved fluorescence, fluorescence homogène en temps résolu) pour mesurer l′activité de la carboxypeptidase B sous un format de criblage miniaturisé haut débit.[1] Elle a été utilisée pour évaluer l′impact de la ou des lysines C-terminales dans la liaison du plasminogène humain à Bifidobacterium. L′effet du traitement par la carboxypeptidase B, une endopeptidase spécifique pour la lysine C-terminale, était mesuré par analyse de cytométrie en flux.[2]
Actions biochimiques/physiologiques
La carboxypeptidase B est une enzyme protéolytique capable d′hydrolyser rapidement les liaisons peptidiques pour libérer certains acides aminés basiques carboxy-terminaux des peptides et protéines. Son poids moléculaire est de 34 300 ± 600 Da. Elle contient un atome-gramme non dialysable de zinc par mole. L′activité enzymatique est inhibée par les agents chélateurs métalliques 1,10-phénanthroline, acide 8-hydroxyquinoline-5-sulfonique et 2,2′-dipyridyle.[3]
