Description générale

Bcl-2-like protein 1 (BCL2L1) or Bcl-2-related gene x, long isoform (Bcl-x) gene is mapped to human chromosome 20q11.21. BCL2L1 belongs to the Bcl-2 family. The bcl-x gene on alternative splicing leads two distinct proteins. It comprises multiple Bcl-2 homology (BH3) domains. Bcl-xL is expressed in the adult brain.(10)

Immunogène

synthetic peptide corresponding to the N-terminal of human Bcl-x (amino acids 2-20) conjugated to BSA. This sequence is identical in human and mouse Bcl-xS, Bcl-xL, Bcl-Xβ and Bcl-Xγ isoforms and is highly conserved in rat Bcl-xS and Bcl-xL (single amino acid substitution).

Application

Anti-Bcl-x antibody produced in rabbit may be used in immunoblotting[1] and immunohistochemistry.[2]

Actions biochimiques/physiologiques

Bcl-2-like protein 1 (BCL2L1) or Bcl-2-related gene x arelong isoforms (Bcl-x) of proteins that are involved in regulating apoptosis. Similar to Bcl-2, Bcl-xL can inhibit cell death upon growth factor removal, when transfected in an IL-3 dependent cell line. Bcl-xL can form ion channels in biological membranes and may maintain cell survival by regulating the permeability of intracellular membranes. Bcl-xL may bind to apoptotic protease activating factor 1 (Apaf-1) preventing it from activating caspase 9. Like Bcl-2, Bcl-x activity is regulated by specific phosphorylation of a 60-amino acid regulatory loop region. Bcl-xL has been shown to regulate cell response to oxidative stress, cellular resistance to DNA damage-induced apoptosis by chemotherapeutic agents and ionizing radiation, and favors neurite outgrowth.

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 1% bovine serum albumin and 15 mM sodium azide.

Stockage et stabilité

For continuous use, store at 2-8 °C for up to one month.For extended storage, freeze in working aliquots.Repeated freezing and thawing is not recommended.Storage in "frost-free" freezers is not recommended. Ifslight turbidity occurs upon prolonged storage, clarifythe solution by centrifugation before use. Workingdilution samples should be discarded if not used within12 hours.
