Description générale

C-C chemokine receptor-like 2 (UniProt: O35457; also known as Chemokine receptor CCR11, G-protein coupled beta chemokine receptor, Lipopolysaccharide-inducible C-C chemokine receptor, L-CCR, CCRL2) is encoded by the Ccrl2 (also known as Ccr11, Lccr) gene (Gene ID: 54199) in murine species. CCRL2 is a seven transmembrane, G-protein coupled receptor that serves as a receptor for CCL19 and chemerin/RARRES2. It does not appear to be a signaling receptor, but may have a role in modulating chemokine-triggered immune responses by capturing and internalizing CCL19 or by presenting RARRES2 ligand to CMKLR1, a functional signaling receptor. CCRL2 is expressed by the majority of cell types of hematopoietic origin, including monocytes, macrophages, neutrophils, lymphocytes, and NK cells. In some cell types, its expression is upregulated by inflammatory stimuli. CCRL2 induction is shown to be dependent on NF-kappaB and JAK/STAT signaling pathways. It is constitutively expressed by mast cells and is detected in bronchial epithelium in OVA-induced airway inflammation. It is reported to be up-regulated during dendritic cell (DC) maturation. It is also considered to be important for the development of Th2 responses and may act as a decoy receptor capturing and endocyting its ligands CCL5 and CCL19. (Ref.: Monnier, J. et al. (2012). J. Immunol. 189(2):956-967).

Spécificité

Clone BZ5B8 reacts with mCCRL2, but not mCMKLR1, huCMKLR1, mCRTH2, huCCRL2, or mCCR10. Clone BZ5B8 immunostained the surface of mCCRL2-expressing cells, but not non-mCCRL2-expressing cells (Zabel, B.A., et al. (2008). J. Exp. Med. 205(10):2207-2220).

Application

Flow Cytometry Analysis: 0.1-100 µg/mL from a representative lot immunostained mouse peritoneal exudate cells from wild-type, but not Ccrl2-/- mice (Courtesy of Brian Zabel, Ph.D., Stanford University, USA).

Flow Cytometry Analysis: A representative lot immunostained L1.2 mouse pre-B lymphoma cells expressing transfected mCCRL2, but not untransfected L1.2 or L1.2 transfectants expressing mCMKLR1, huCMKLR1, mCRTH2, huCCRL2, or mCCR10, nor non-CCRL2-expressing mouse blood cells. Chemerin, but not CCL2, blocked clone BZ5B8 staining of CCRL2-positive mouse peritoneal exudate cells (Zabel, B.A., et al. (2008). J. Exp. Med. 205(10):2207-2220).
