Gene Information

human ... CASP9(842)

General description

CASP9 (Caspase-9) gene encodes a protein belonging to the cysteine-aspartic acid protease (caspase) family. It consists of two domains. The large subunit of caspase 9 possesses a seven-residue loop at the C terminal end. The loop helps to interact across the dimer interface.

Immunogen

Caspase-9 precursor recombinant protein epitope signature tag (PrEST)

Application

Anti-CASP9 antibody produced in rabbit, a Prestige Antibody, is developed and validated by the Human Protein Atlas (HPA) project . Each antibody is tested by immunohistochemistry against hundreds of normal and disease tissues. These images can be viewed on the Human Protein Atlas (HPA) site by clicking on the Image Gallery link. The antibodies are also tested using immunofluorescence and western blotting. To view these protocols and other useful information about Prestige Antibodies and the HPA, visit sigma.com/prestige.
Applications in which this antibody has been used successfully, and the associated peer-reviewed papers, are given below.

Biochem/physiol Actions

CASP9 (Caspase-9) is involved in the apoptotic pathway. The caspases exist as inactive proenzymes that are activated upon proteolytic processing at conserved aspartic residues. The active enzyme is a dimer of one small and one large subunit. It functions as an initiator caspase that is activated by mitochondrial-mediated apoptotic pathway. It executes apoptosis by activating a cascade of effector caspases. It also acts as a tumor suppressor.

Features and Benefits

Prestige Antibodies® are highly characterized and extensively validated antibodies with the added benefit of all available characterization data for each target being accessible via the Human Protein Atlas portal linked just below the product name at the top of this page. The uniqueness and low cross-reactivity of the Prestige Antibodies® to other proteins are due to a thorough selection of antigen regions, affinity purification, and stringent selection. Prestige antigen controls are available for every corresponding Prestige Antibody and can be found in the linkage section.

Every Prestige Antibody is tested in the following ways:
  • IHC tissue array of 44 normal human tissues and 20 of the most common cancer type tissues.
  • Protein array of 364 human recombinant protein fragments.

The ability of p53 to promote apoptosis in response to mitogenic oncogenes appears to be critical for its tumor suppressor function. Caspase-9 and its cofactor Apaf-1 were found to be essential downstream components of p53 in Myc-induced apoptosis. Like p53
