General description

Alcian blues, are a family of polyvalent basic dyes, of which, the Alcian blue 8G has been historically the most common and the most reliable member. It is used to stain acidic polysaccharides such as glycosaminoglycans in cartilages and other body structures, some types of mucopolysaccharides, sialylated glycocalyx of cells etc. Alcian blue if often used to identify Chondrocytes from differentiated cultures of both human and rodent mesenchymal stem cells.

Questions

1–4 of 4 Questions
  1. What is the composition of the differentiation for TMS 010 C procedure?

    1 answer
    1. Product TMS-010 is a standard pH 2.5 solution of Alcian Blue that stains a number of substances, including chondrocytes, at this pH. The provided procedure is similar but not identical to the standard histology stain procedure found in most histology textbooks.

      Helpful?

  2. What is the concentration of acetic acid specified for the Alcian Blue pH 2.5 TMS-010-C product?

    1 answer
    1. The concentration of acetic acid specified for the Alcian Blue pH 2.5 TMS-010-C product is not provided. However, it is a classical textbook formulation typically consisting of 1% Alcian Blue in 3% glacial acetic acid.

      Helpful?

  3. Is there a procedure available for using TMS-010, and in this context, would you anticipate the alcian blue to be present in the nuclei, cytoplasm, or the entire cell?

    1 answer
    1. Depending on the preparation of Alcian Blue, the dye can be used to identify various substances. TMS-010 seems to have been formulated to stain chondrocytes, the cells that produce collagen. It's expected that the staining should be primarily in the cytoplasm. However, if the staining intensity is high, it might appear that the entire cell is stained.

      Helpful?

  4. Can you provide a protocol for a colorimetric assay that uses Alcian Blue to compare sulfated proteoglycan content in adherent cancer cell lines?

    1 answer
    1. The Alcian-Blue Staining Solution (cat. no. TMS-010-C) is a cell culture product from the cell culture portfolio, intended for cell-based assays, cell differentiation, and stem cell culture. For example, to identify chondrocytes from differentiated cultures of both human and rodent mesenchymal stem cells (MSCs).

This product is presented as a 1% w/v Alcian Blue solution in acetic acid at pH 2.5.

The Certificate of Analysis does not include any usage instructions and there is no separate product data sheet or user guide.
