General description

Protein-arginine deiminase type-4 (PAD4; formerly PAD5) is one of five PAD (1-4, 6) homologues that belong to the superfamily of multidomain amidinotransferases. The PAD enzymes catalyze deiminating reactions in which guanidinium is hydrolyzed to peptidylcitrulline. PAD4 is a calcium-regulated nuclear-localized enzyme. It modifies arginine residues on histones H2A, H3, and H4 (H2 Arg3; H3 Arg2, H3 Arg8, H3 Arg17, H3 Arg26; and H4 Arg3). Several studies have suggested that PAD4-mediated citrullination has important implications for gene regulation. PAD4 may perform deimination at sites overlapping with arginine methylation, and antagonize the transcription-promoting effects of methylating enzymes such as CARM1 and PRMT1. PAD4 may therefore inhibit gene expression, most notably expression of cell-cycle regulating p53 genes such as p21. Previous studies have suggested that PAD4 may play central role in the development of cancer.
