WNK3 is a serine-threonine protein kinase and expressed throughout the nephron, but most commonly at intercellular junctions. In its active state, it is a promoter of NKCC2 and NCC activity. The opposite is true of WNK3 when in an inactive state. WNK3 changes the expression of these transporters in the membrane and is thought to be a regulator of blood pressure along with other members of the WNK family of protein kinases. In recent research, WNK3 has also been linked to regulating signaling cascades involved in tumorigenesis.
