General description

α-amylase is the main secretory protein constituent of saliva. It contains 496 amino acids and has two isoforms, glycosylated and non-glycosylated.[1] α-amylase gene is located on human chromosome 1p21.1.[2] α-amylase is mainly produced in pancreas and saliva.[3]

Application

α-amylase from human saliva has been used to test α-amylase inhibitors using HPLC (high performance liquid chromatography).[4] It also has been used to study the digestibility of human milk oligosaccharides (HMO).[5]

Biochem/physiol Actions

α-amylase catalyses the hydrolysis of α-1,4 glycosidic linkage in oligosaccharides. It plays a crucial role in the initial digestion of starch, glycogen and polysaccharides.[1] Elevated levels of amylase in serum is linked to acute pancreatitis.[3]

Unit Definition

One unit will liberate 1.0 mg of maltose from starch in 3 min at pH 6.9 at 20 °C.

  1. Good morning, I allow myself to contact you to know the mass of the product "α-amylase from Human Saliva" (A0521-100UN). It lacks the mass of the product and I would like to know it.

    1. The actual amount of powder per pack will depend on the units/mg protein of the lot received. This value will be provided in the lot-specific Certificate of Analysis. For example, lot SLCN0121 tested at 1474 units/mg protein. The 100UN pack size will provide 0.06mg of protein, the 500UN pack size will provide 0.34mg of protein, and the 2500UN pack size will provide 1.70mg of protein.   2. Dear Dr, how much mg saliva amylase are present in glass. We design the experimet sets. So need to know the Total amount of the amylase(mg) in the bottle. Thanks

    1. The exact amount is determined by the units/mg protein listed on the Certificate of Analysis. For example, the lot SLCN0121 tested at 1474 units per milligram protein. The total amount of the amylase (mg) in the bottle for the 100 unit package contains 0.07 mg, the 500 unit package contains 0.34 mg, and the 2500 unit package will contain 1.70 mg of protein. This lot is 11.8% protein. These values will change from lot to lot, but the actual mass in the bottles remains low. Other products, such as A4551, are sold by weight.
