Specificity

Clone 3B9 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects human IL-6.

Immunogen

His-tagged recombinant full length human IL-6 (without the signal peptide).

Application

Quality Control Testing

Evaluated by Western Blotting in human recombinant IL-6.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected human recombinant IL-6.

Tested applications

Affinity Binding Assay Analysis: A representative lot of this antibody bound IL-6 with a KD of 1.0 x 10^-12 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected IL-6 in K562 cell line.

ELISA Analysis: A representative lot of this antibody detected human recombinant IL-6 in ELISA application.

Anti-IL-6, clone 3B9 ZooMAb®, Cat. No. ZRB1982, is a recombinant Rabbit monoclonal antibody that specifically targetsIL-6 and is tested for use in Affinity Binding Assay, ELISA, Immunocytochemistry, and Western Blotting.

Target description

Interleukin-6 (UniProt: P05231; also known as IL-6, B-cell stimulatory factor 2, BSF-2, CTL differentiation factor, CDF, Hybridoma growth factor, Interferon beta-2, IFN-beta-2) is encoded by the IL6 (also known as IFNB2) gene (Gene ID: 3569) in human. IL-6 is a proinflammatory cytokine that is synthesized with a signal peptide (aa 1-29), which is subsequently cleaved off to generate the mature form. It is produced by monocytes, endothelial cells, and adipose tissue. IL-6 is involved in a variety of biological functions and is a potent inducer of acute phase response. In the innate immune response. It is synthesized by myeloid cells, such as macrophages and dendritic cells, upon recognition of pathogens through toll-like receptors (TLRs) at the site of infection or tissue injury. However, in the adaptive immune response, it is required for the differentiation of B cells into immunoglobulin-secreting cells. It also plays a major role in the differentiation of CD4+ T cell subsets and in proliferation of myeloma cells and the survival of plasma blast cells. It drives naive CD4+ T cells to the Th17 lineage. IL-6 is reported to discharge into the bloodstream after muscle contraction and increase lipolysis and improves insulin resistance. Rapid production of IL-6 contributes to host defense during infection and tissue injury, but excessive IL-6 synthesis contributes to disease pathology. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
