Specificity

Clone 1F16 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects Guanine nucleotide-binding protein subunit alpha-11 (Ga 11). It targets an epitope within 10 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 10 amino acids from the C-terminal region of human Guanine nucleotide-binding protein subunit alpha-11 (Ga 11).

Application

Anti-GNA-11 (G 11), clone 1F16 ZooMAb, Cat. No. ZRB1446, is a recombinant Rabbit monoclonal antibody that specifically targets GNA-11 (G protein alpha 11) and is tested in Immunocytochemistry and Immunohistochemistry (Paraffin), and Western Blotting.
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected GNA-11 (Ga 11) in HeLa, NIH3T3, and rat brain microsomal lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected GNA-11 (Ga 11) in human placenta tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected GNA-11 (Ga 11) in HeLa, A431, HUVEC and NIH 3T3 cells.

Target description

Guanine nucleotide-binding protein subunit alpha-11(UniProt: P29992; also known as G alpha-11, G-protein subunit alpha-11, Guaninenucleotide-binding protein G(y) subunit alpha) is encoded by the GNA11 (alsoknown as GA11) gene (Gene ID: 2767) in human. G proteins are composed of 3units; alpha (36-52 kDa), beta (35-36 kDa), and gamma (8-10 kDa). The alphachain contains the guanine nucleotide binding site. When bound to GDP, G alpha associates withthe beta-gamma dimer to form the inactive heterotrimer. Ligand/agonist bindingto a GPCR alters the receptor’s conformation allowing it to catalyze theexchange of GDP for GTP on the alpha subunit of the G protein coupled to thereceptor. The binding of GTP produces aconformational change in the G protein alpha subunit resulting in thedissociation of the receptor-G protein complex and freeing the alpha andbeta-gamma subunits to interact with their target effectors. The slow intrinsic GTPase activity of thesubunit hydrolyzes the bound GTP to GDP ending the interaction of the subunitwith its’ effector. The GDP bound alphasubunit interacts strongly with beta-gamma subunits and thus GTP hydrolysispresumably also terminates beta-gamma mediated signaling by promoting theassociation of the GDP-bound subunits with beta-gamma dimers. The G alpha 11 targets phospholipase C (PLC)that cleaves phosphatidylinositol 4,5-bisphosphate [PtdIns(4,5)P2] to generateinositol trisphosphate (IP3) and membrane bound diacylglycerol (DAG). ThisZooMAb® recombinant monoclonal antibody, generated by our propriety technology,offers significantly enhanced specificity, affinity, reproducibility, andstability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in lyophilized from PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

0.3 mg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
