Application

BME is suitable for reducing protein disulfide bonds prior to polyacrylamide gel electrophoresis and is usually included in a sample buffer for SDS-PAGE at a concentration of 5%. Cleaving intermolecular (between subunits) disulfide bonds allows the subunits of a protein to separate independently on SDS-PAGE. Cleaving intramolecular (within subunit) disulfide bonds allows the subunits to become completely denatured so that each peptide migrates according to its chain length with no influence due to secondary structure.
Reducing agent
β-mercaptoethanol or 2-mercaptoethanol has been used:
  • as a supplement in Roswell Park Memorial Institute (RPMI)-1640 medium to culture DT40 cells (chicken B cell line)[1]
  • to lyse NuLi-1 (Normal Lung, University of Iowa-1) cells[2]
  • to trypsinize mouse embryonic stem cells[3]

  1. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.   3. I’ve purchased 25mL bottle and want to use this in my cell culture media at concentration of 50mM. What is the concentration in the 25mL bottle and how to make this to 50mM concentration?

    1. This product is a neat or pure material. The concentration is considered to be 100%. The calculated molarity is 14.3 M as reported in the Properties section of the product page.
      To prepare a 50 mM solution, please mix 0.391 g of M3148 with 100 mL solvent. 0.391 g / 78.13 g/mol / 0.1 L = 0.05 = 50 mM.
