General description

Programmed cell death protein ligand 1 (PD-L1), also known as cluster of differentiation 274 (CD274) and B7-H1, is a type 1 membrane protein of 290 amino acids. It is found in a variety of cell types and binds programmed cell death protein (PD-1). It has been indicated as playing a role in autoimmune disease, allograft responses, pregnancy, and other disease states. When PD-L1 binds to PD-1, an inhibitory signal is transmitted that ultimately reduces the proliferation of antigen-specific T cells and reduces the rate of apoptosis in regulatory T-cells. By doing so, PD-L1 contributes to the immune evasion of cancer tissues by suppressing the adaptive immune system. Blocking this activity is thought to restore anti-tumor activity. High PD-L1 expression is correlated with poor prognosis with some cancer types. High levels of the soluble form are also associated with poor prognosis and advanced tumor progression. Multiple PD-L1 inhibiting monoclonal antibodies have been approved as cancer therapeutics.
