General description

Morphine-6-β-D-glucuronide is a primary plasma and urinary metabolite of the opiate analgesic morphine. Sold under trade names including MS Contin®, Kadian®, Oramorph®, and Roxanol for pain relief, morphine is also a commonly abused recreational drug. This certified analytical reference standard is suitable for a variety of GC/MS or LC/MS applications from pain prescription monitoring and urine drug testing to clinical toxicology and forensic analysis.

Application

  • Pharmacokinetics and Clinical Study Applications: A method involving liquid chromatography coupled to tandem mass spectrometry was developed and validated for the simultaneous quantification of several analgesics, including Morphine-6-β-D-glucuronide, applied in a clinical study for determining neurological death in intensive care units (Jutras et al., 2020).
  • Opioid Receptor Research: Research highlighted the higher efficacy of Morphine-6-β-D-glucuronide compared to morphine as a mu-opioid receptor agonist in the rat locus coeruleus, supporting its use in detailed opioid receptor studies (Osborne et al., 2000).

Synthesis and biological evaluation of new derivatives of Morphine-6-Glucuronide (M6G) are described. M6G is an active metabolite of morphine which displays more analgesia than morphine with a superior side effect profile but with a less efficiently BBB penetration. These phenomena

Opioid analgesics exhibit cationic properties under physiological conditions, and the mechanism underlying permeation of the blood-brain barrier thus cannot be fully explained by simple diffusion alone. Various types of transporters that exhibit substrate specificity are localized on the blood-brain barrier

Interindividual variability in analgesic response and adverse effects of opioids because of narrow therapeutic indices are major clinical problems. Morphine is an opioid commonly used in children to manage perioperative pain. Al-though size and age often are considered primary covariates
