General description

1-phosphatidylinositol 4,5-bisphosphate phosphodiesterase eta-2 (UniProt: O75038; also known as EC:3.1.4.11, Phosphoinositide phospholipase C-eta-2, Phosphoinositide phospholipase C-like 4, PLC-L4, Phospholipase C-like protein 4, Phospholipase C-eta-2, PLC-eta2) is encoded by the PLCH2 (also known as KIAA0450, PLCL4) gene (Gene ID: 9651) in human. PLCH2 is a member of the PLC-eta family of the phosphoinositide-specific phospholipase C (PLC) superfamily of enzymes that cleave PtdIns(4,5) P2 to generate second messengers inositol 1,4,5-trisphosphate (IP3) and diacylglycerol (DAG). Its expression is observed in retina and kidney. It may be involved in the formation and maintenance of neural network in the postnatal brain. Its phospholipase activity is shown to be very sensitive to calcium. Its PH domain is localized in amino acids 47-155 and its N-terminal region, comprising amino acids 1-155, is necessary for its plasma membrane localization. At its C-terminus it has a class II PDZ-binding motif. PLCH2 activity is reported to be directly regulated by β G-protein subunits. Five isoforms of PLCH2 have been described that are produced by alternative splicing. Quality Control Testing

Evaluated by Western Blotting with His-ABP-tagged recombinant fragment of Human PLCH2.

Western Blotting Analysis (WB): A 1:10,000 dilution of this antibody detected His-ABP-tagged recombinant fragment of Human PLCH2.

Tested Applications

Enzyme Immunoassay (ELISA) Analysis: Various dilutions of this antibody detected recombinant fragment of human PLCH2.

Affinity Binding Assay: A representative lot of this antibody bound PLCH2 peptide with a KD of 5.4 x 10-7 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 3J3 is a ZooMAb^® Rabbit recombinant monoclonal antibody that specifically detects PLCH2. It targets an epitope within 21 amino acids from the internal region.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
