Application
Endoproteinase Glu-C aus Staphylococcus aureus V8 wurde wie folgt verwendet:
- Zum Erhalt proteolytischer Spaltungsfragmente des S-Schicht-Proteins (aus Bacillus stearothermophilus ATCC 12980) zur Durchführung von Affinitätsstudien.[1]
- Zur enzymatischen Spaltung von nativem VSTx-3 (Voltage Sensor Toxin 3)-Peptid zur Bestimmung seiner Sequenz.
- Zur eingeschränkten Proteolyse von rekombinanter aufgereinigter PimA (Phosphatidylinositolmannosyltransferase).
- Beim Aufschluss von glykolysiertem Hämoglobin zur Isotopenverdünnung-Flüssigchromatographie-Tandem-Massenspektrometrie-Analyse.
Biochem/physiol Actions
Endoproteinase Glu-C aus dem Staphylococcus aures-Stamm V8 ist eine Serinendoprotease, die Peptidbindungen an der Carboxyseite der Glutamyl- und Aspartylreste hydrolysiert. Die Spezifizität von Glu-C ist von dem eingesetzten Puffer und pH-Wert sowie der Struktur um die potenzielle Spaltungsstelle herum abhängig. In Ammoniumacetat (pH-Wert 4,0) oder Ammoniumbicarbonat (pH-Wert 7,8) spaltet das Enzym bevorzugt Glutamylbindungen, während Glu-C in Phosphatpuffer (pH-Wert 7,8) an jeder Stelle spaltet. Glu-C ist in Gegenwart von 0,2 % SDS (Natriumdodecylsulfat) und in 4,0 M Harnstoff aktiv.
