Allgemeine Beschreibung

Platelet-Derived Growth Factor (PDGF) is a cationic polypeptide and a constituent of α-granules. PDGF is composed of chain A and B linked together by a disulphide bridge.[1] PDGF heterodimers occurs as PDGF-AB and homodimers exists as FDGF-AA, PDGF-BB, PDGF-CC and PDGF-DD).[2] The PDGFB gene is localized to pericytes in the brain and is mapped to human chromosome 7p22.3.[3][4]

Anwendung

Platelet-Derived Growth Factor-AB human has been used in luciferase reporter assay.[5]

Biochem./physiol. Wirkung

Cellular actions of platelet-derived growth factor (PDGF) include chemoattraction and activation of neutrophils, monocytes, and fibroblasts.[6] PDGF appears to play a crucial role in the cellular response to tissue injury and is critically involved in the pathological progression of atherosclerosis.
PDGF stimulates fibroblast proliferation and extracellular matrix production in order to facilitate tissue repair.[7] PDGF-AB isomer is found to be associated with angiogenesis and proliferation.[8] Loss of PDGFB gene function induces microvascular calcification due to age-dependent phenotypical changes.[3]

Sonstige Hinweise

Heterodimer of the A and B chains of PDGF disulfide linked.

Physikalische Form

Lyophilized from a 0.2 μm filtered solution in 40% acetonitrile and 0.1% trifluoroacetic acid.

Hinweis zur Analyse

The biological activity is measured by its ability to stimulate 3H-thymidine incorporation in quiescent NR6R-3T3 fibroblasts.
