General description
Pertussis-Toxin (PTX) ist ein Protein, das aus dem Bakterium Bordetella coccobacillus isoliert wird. Das Toxin wird in einem nicht aktiven Zustand ausgeschieden. Wenn das B-Oligomer des Bakteriums Bordetella coccobacillus in Kontakt mit der Zellmembran kommt, wird eine S1-Untereinheit seines A-Protomers aktiviert. Diese Aktivierung kann durch Glutathion und/oder ATP vermittelt werden.
Application
Das Pertussis-Toxin von Bordetella pertussis wird wie folgt verwendet: zur Verstärkung der kardialen Myosin-Immunisierung von Ratten zur Induktion einer Myokarditis[1], zum Aufbrechen der Blut-Hirn-Schranke im Cuprizon- und Cuprizon-/MOG-EAE-Modell[2], als Inhibitor der G-αi-Signalübertragung[3]
Das Toxin wird von B. pertussis in einer inaktiven Form freigesetzt. Wenn das B-Oligomer des Pertussistoxins an die Zellmembran bindet, wird die S1-Untereinheit seines A-Protomers möglicherweise durch die Wirkung von Glyutathion und ATP aktiviert. Ein Protokoll für die Aktivierung des Pertussistoxins in vitro wird von Kaslow, et al vorgegeben.

Das Pertussistoxin katalysiert die ADP-Ribosylierung der α-Untereinheiten der heterotrimeren Guanin-Nukleotid-Regulatorproteine Gi, Go und Gt.

Dadurch wird die Wechselwirkung der G-Protein-Heterotrimere mit Rezeptoren verhindert und deren Kupplung und Aktivierung blockiert. Da die Gα-Untereinheiten in ihrem GDP-gebundenen, inaktiven Zustand verbleiben, sind sie nicht in der Lage, Adenylylcyclase zu inaktivieren oder K+-Kanäle zu öffnen.
