General description

Cholesterin ist ein wichtiger Baustein mammalischer Zellmembranen, wo es eine Rolle beim intrazellulären Transport, beim Senden der Zellsignale und der Erhaltung der Membranfluidität spielt. Im Blut zirkuliert Cholesterin sowohl als freie Säure wie auch als Cholesterin-Ester. Die Kontrolle von Cholesterin im Serum spielt eine wichtige therapeutische Rolle, da erhöhte Cholesterinspiegel mit der Entwicklung von Atherosklerose und kardiovaskulären Pathologien in Zusammenhang stehen. Jüngste Erkenntnisse deuten darauf hin, dass eine Störung der Cholesterinhomöostase zur Entwicklung eines Zustandes chronischer Entzündung führt.

Cholesterin wird vorwiegend in der Leber produziert. Anormale Cholesterinspiegel werden mit koronarer Herzkrankheit in Verbindung gebracht. Cholesterin steht in Zusammenhang mit dem Fortschreiten oder der Hemmung von Brustkrebs. Tumorzellen sind in der Lage, ihr eigenes Cholesterin zu synthetisieren, was die Zellproliferation fördert. Ein erhöhter Cholesterinspiegel wurde bei Krebserkrankungen beobachtet, insbesondere bei Prostatakrebs. Erhöhte Cholesterinwerte im Serum wurden bei Fettleibigkeit festgestellt.

Application

Das Cholesterin-Quantifizierungskit wurde für die Messung der Gesamtcholesterinkonzentration im Serum verwendet.
Zur Verwendung mit Zell- und Gewebekulturproben, Urin, Plasma, Serum und anderen biologischen Proben geeignet.

Biochem/physiol Actions

Das Cholesterin-Quantifizierungskit lässt sich auch verwenden, um die Konzentration von in einer Probe vorhandenem freiem Cholesterin, von Cholesterinester oder beidem (Gesamtwert) zu bestimmen. In diesem Kit wird die Cholesterinkonzentration durch einen gekoppelten Enzym-Assay bestimmt, der ein kolometrisches (570 nm)/fluorometrisches (λex = 535/λem = 587 nm) Produkt ergibt, das proportional zum vorhandenen Cholesterin ist.

  1. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  2. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.   3. Hi, Can the cholesterol quantification assay (MAK043) measure the cholesterol content in lipid nanoparticles? Does it break down the particles and measure total cholesterol or does it measure only unreacted cholesterol?

    1. The Cholesterol Quantitation Kit can also determine the concentration of free cholesterol, cholesteryl esters, or both (total) present in a sample. The assay is enzyme-based and needs the cholesterol or cholesterol esters in solution rather than in a membrane type structure. The kit has not been tested directly for use with lipid nanoparticles.

  4. Can you use this kit on frozen animal cell pellets?

    1. It is recommended to use fresh samples for this kit. However, although not ideal, flash-frozen tissue or cells stored at -80C can be used. It is important to remember that results may vary between fresh and frozen samples. If possible, fresh and frozen samples should be run parallel to compare results.

  5. Can this kit be used with bacterial supernatant (cultivation medium without cells) and how prepare these samples? Is necessary organic extraction?

    1. This product has not been tested for use in bacterial culture supernatent, however there is no reason to suspect that this kit would not be suitable. Solid tissues and cell lysates are extracted using chloroform:isopropanol:Igepal preparation. Samples such as serum, plasma, or other biological fluids do not require organic extraction. The minimum concentration for detection has not been determined, although the first data point in the standard curve is 1 ug/well for the colorimetric assay and 0.1 ug/well for the fluorometric assay.   6. What's the limit of detection (LOD) and detection range for cholesterol quantitation kit?

    1. The absolute limit of detection of this kit is not determined. However the kit is able to quantify at least 1 ug/well (colorimetric) or 0.1 ug/well (fluorimetric).

  7. Hi, we need to quantify cholesterol in shrimps mescle samples, are possible?

  10. Can these kits be used with serum samples from mouse?

    1. Yes, this kit can be used on mouse serum samples. We generally recommend that fresh serum samples be used. Frozen samples can probably be used. However, the successful use of frozen samples depends on the efficiency of freezing them and the amount of time they have been frozen. If previously frozen serum samples are used, you may want to check the performance of the kit with fresh vs. frozen samples.
