General description

Fluoromount ist, ähnlich wie Gel Mount, ein wasserbasiertes Eindeckmediumä und wird zusammen mit einem Deckglas zur Erhaltung von mit fluoreszierenden Farbstoffen gefärbten Geweben verwendet. Dieses Produkt ist kompatibel mit den Markern FITC, Phycoerythrin, Phycocyanin und Allophycocyanin. Darüber hinaus kann es auch mit anderen fluoreszierenden Markern wie TexasrotÒ und Rhodamin verwendet werden. Das Medium kann nach der Verwendung leicht entfernt werden, indem die Objektträger in entionisiertes Wasser getaucht werden.

Application

Fluoromount™ wässriges Fixiermittel wird verwendet, um folgendes zu fixieren:
  • Deckgläser auf Traggläsern zur Immunfluoreszenz-Mikroskopuntersuchung von PC12- und Hela-Zellen
  • Hippocampus-Neuronenzellen für Konfokalmikroskopuntersuchungen
  • Gehirnschnitte zur Immunfluoreszenzfärbung[1]

    1. This product is provided as a clear, viscous solution. Refrigeration may cause it to become difficult to use for coverslipping. Its product Information states, "Do not refrigerate". This applies to storage after the product has been opened.

  2. What are some examples of aqueous mounting media without glycerol?

    1. For use with fluorescence:

      1. F4680 Fluoromount
      2. F6182 Fluoroshield

      Aqueous Mounting media with no glycerol and antifade agents:

      1. 10981 Polyvinyl alcohol with DABCO
      2. F6937 Fluoroshield with DABCO
      3. F6812 Fluoroshield with PPD (not suitable for a select number of fluorescent dyes)

      Aqueous Mounting Media with no glycerol and another dye:

  3. Is F4680 suitable for use as a mounting medium for AEC red IHC mounting?

    1. Fluoromount is not intended for use with bright field microscopy or for coverslipping slides stained with AEC. Most fluorescent mounting media are somewhat alkaline, with a pH around 9, while AEC is typically formulated with an acetate buffer around pH 5.2. The difference in pH levels suggests that the AEC may fade quickly, especially if the slides are counterstained with a nuclear counterstain such as Mayer's Hematoxylin, which would also likely disappear rapidly.

  4. Does this (Fluoromount F4680) contain DAPI?

    1. This product does not contain DAPI. Should DAPI be preferred, see product F6057.

  5. How does the storage temperature relate to shipping conditions?

    1. The storage conditions that a Sigma-Aldrich catalog and label recommend for products are deliberately conservative. For many products, long-term storage at low temperatures will increase the time during which they are expected to remain in specification and therefore are labeled accordingly. Where short-term storage, shipping time frame, or exposure to conditions other than those recommended for long-term storage will not affect product quality, Sigma-Aldrich will ship at ambient temperature. The products sensitive to short-term exposure to conditions other than their recommended long-term storage are shipped on wet or dry ice. Ambient temperature shipping helps to control shipping costs for our customers. At any time, our customers can request wet- or dry-ice shipment, but the special handling is at customer expense if our product history indicates that the product is stable for regular shipment.

  6. Does Fluoromount mounting media (Product No. F4680) contain an antifade reagent?

    1. Fluoromount does not contain an antifade reagent.  If you need an antifade mounting media, check Fluoroshield (Product Nos. F6182, F6057 and F5932) or PVA with antifade (Product Nos. 10981 and 10979).

  7. How do you remove the coverslip after using Fluoromount (Product No. F4680)?

    1. Place the slide in a beaker full of deionized water on a magnetic stirrer. Gently stir the slide for a few hours to overnight for complete removal of dry Fluoromount.

    1. Product No. F4608, Fluoromount is essential the same as GelMount. GelMount was deleted by the supplier. Fluoromount is being made by another supplier with the same formulation.

    1. Fluoromount does not contain glycerol. The exact formulation is considered proprietary.
