General description
Application
Exendin-4 wurde für die Behandlung des ob/ob-Mausmodells zur Bestimmung der Glucose- und Insulinkonzentrationen sowie für ein Homeostase-Bewertungsmodell (HOMA)[4] und zur Bestimmung des Nuklearfaktor-κΒ (NF-κB) p65-Spiegels in mit Exendin-4 behandelten Makrophagen verwendet.[5]
Exendin-4 wurde als Inkretin-Mimetikum-Behandlung in HepG2-Zellen verwendet, um den Einfluss von Hilfsstoffen auszuschließen. Es wurde auch als r (GLP-1R)-Agonist für den Ausschluss von Hilfsstoff-Interferenzen verwendet.
Biochem/physiol Actions
Es aktiviert GLP-1(Glucagon-ähnliche Peptid-1)-Rezeptoren für eine Erhöhung des intrazellulären cAMP-Spiegels in pankreatischen azinären Zellen; es hat keine Wirkung auf VIP-Rezeptoren.
Exendin-4 imitiert die Aktivität des Inkretinhormons Glucagon-like Peptid 1 (GLP-1) und wirkt somit bei der Kontrolle der Glucose mit. Zu diesen Wirkungen gehören die Sekretion von Insulin auf glucoseabhängige Weise, negative Regulation einer hohen Glucagonsekretion und verlängerte Dauer der Magenentleerung. Bei Patienten mit Typ-2-Diabetes kann dieses Peptid für die glykämische Kontrolle subkutan verabreicht werden, wenn mit Metformin keine ausreichenden Ergebnisse erzielt werden.[1] Es fördert die Neogeneration und Proliferation von β-Zellen und unterstützt somit die Regeneration der Pankreas.[2] Es dient als Ligand für den Exendin-Rezeptors und führt zu einer Erhöhung des cAMP-Spiegels in azinären Zellen.[3]
