General description

Avidin ist ein tetrameres oder dimeres Biotin-bindendes Protein, das in den Eileitern von Vögeln, Reptilien und Amphibien gebildet wird und im Eiweiß (Eiklar) ihrer Eier enthalten ist.[1] Es besteht aus vier hochaffinen Bindungsstellen für Biotin. ExtrAvidin® wird aus dem im Eiklar enthaltenen Avidin hergestellt. Es ist eine modifizierte Form von affinitätsbereinigtem Avidin mit der hohen spezifischen Aktivität von Avidin und der geringen Hintergrundanfärbung von Streptavidin. Dies ist ein Biotin-bindendes Protein, das von dem Bakterium Streptomyces avidinii [2]produziert wird. ExtrAvidin wurde mit alkalischer Phosphatase konjugiert und wird für verschiedene Verfahren eingesetzt.

Application

ExtrAvidin® – Alkalische Phosphatase wird verwendet in:
  • Dotblot
  • Immunhistochemie (Formalin-fixierte, in Paraffin eingebettete Gewebeschnitte)
  • Indirekter ELISA
  • Enzymgekoppelter Immunabsorptionsassay (ELISA)[3][4]
  • ELISPOT (Enzyme Linked Immunosorbent Spot)-Analyse[5]
  • Kompetitiver ELISA[6]

Biochem/physiol Actions

Alkalische Phosphatase katalysiert die Hydrolyse von Estern zur Gewinnung von Phosphorsäure. Sie erleichtert auch den Transport von Substanzen über Membranen.[7]

Physical form

Lösung in 0,05 M Tris-HCl-Puffer, pH 8,0; enthält 1 mM MgCl2, 1% Rinderserumalbumin und 15 mM Natriumazid
