Das ECM-Gel polymerisiert, wenn es auf 20–40 °C erwärmt wird, und bildet eine rekonstituierte Basalmembran. Bei Kultivierung auf diesem ECM-Gel bilden PC12-Zellen innerhalb von 2 Tagen Neuriten aus.
Jede Mauskolonie, die für die Produktion dieses Produkts verwendet wird, wird routinemäßig auf mehrere Pathogene untersucht. Mit negativem Ergebnis auf LDEV getestet.

Preparation Note
Dieses Produkt wird aus Engelbreth-Holm-Swarm-Sarkom der Maus hergestellt; gegen Chloroform dialysiert. Das Gel sollte vor der Verwendung übernacht bei 2-8 °C aufgetaut und in eine Multiwellplatte pipettiert werden (Platte und Pipetten auf 2-8 °C vorgekühlt). Das Gel kann auch vor der Zugabe zur Platte mit Dulbeccos modifiziertem Eagle-Medium von 2-8 °C bis zu zweifach verdünnt werden. Das Produkt geliert innerhalb von 5 Minuten bei 20 °C. Eine längere Handhabung sollte bei unter 10 °C erfolgen. Die Zellen können auf einer dünnen Gelschicht von 0,5 mm ausplattiert oder in einer 1 mm dicken Schicht kultiviert werden. Wenn Zellen innerhalb der Schicht kultiviert werden, geben Sie die Zellen vor dem Ausplattieren bei der empfohlenen Zelldichte von 3-4 x 104 Zellen pro ml zu. Um Zellen von diesem Gel zu trennen, verwenden Sie Protease mit PBS ohne Calcium, Magnesium oder EDTA bei einer Konzentration von 0,6-2,4 Einheiten pro ml.

ECM wurde auf eine Proteinkonzentration von 8-12 mg/ml (in DMEM) eingestellt und enthält Laminin als Hauptbestandteil sowie Typ-IV-Collagen, Heparinsulfatproteoglycan, Entactin und andere Nebenbestandteile. Die Zugabe von Typ-IV-Collagen zum ECM-Gel verstärkt die Polymerisation.

Dispase (Product No. D4818) is a Bacillus-derived neutral protease that is recommended for recovering cells cultured on ECM (Product No. E1270) or Matrigel. Dispase will yield a single cell suspension far more effectively than trypsin, collagenase, or other proteolytic enzymes. Dispase is gentle and will not damage cells harvested for sub-cultivation or other study.
