Application

Aggrecan ist eine kritische Komponente der Knorpelstruktur und der Funktion von Gelenken. Die Rollen seiner Synthese und seines Abbaus beim Knorpelrückgang bei Gelenksverletzungen und beim Altern werden untersucht.

Biochem/physiol Actions

Das Molekül erzeugt ein rigides, reversibel verformbares Gel, das Kompressionen standhält. In Kombination mit Hyaluronsäure bildet es sehr große Makromolekülkomplexe, die ein vergrößertes hydrodynamisches Volumen und eine signifikante Steigerung (30–40%) der relativen Viskosität der Lösung aufweisen.

Preparation Note

Dieses Produkt wird aus Gelenkknorpeln gewonnen, chromatographisch aufgereinigt, gegen Wasser dialysiert und vor der Lyophilisierung durch einen 0,2-μm-Filter sterilfiltriert. Nach der Lyophilisierung ist dieses Pulver im Wesentlichen salzfrei. Dieses Produkt ist zu 2 mg/ml wasserlöslich.

Aggrecan ist eine wichtiges strukturelles Proteoglykan der extrazellulären Knorpelmatrix. Es ist ein großes Proteoglykan mit einer Molekülmasse von mehr als 2.500 kDa, das aus etwa 100–150 Glykosaminglykanketten, die an ein Kernprotein gebunden sind, besteht. Der Großteil dieser Glykosaminglykanketten ist Chondroitin-/Dermatansulfat. Aggrecan umfasst drei globuläre Domänen, G1, G2 und G3, die an der Aggregation und Hyaluronanbindung, Zellanhaftung und Chondrozytenapoptose beteiligt sind.
