Beschreibung

General description

Guanidiniumchlorid (GdmCl) ist ein bekanntes Protein- und Enzym-Denaturierungsmittel.[1][2] Es ist ein kleines Molekül und von Natur aus hydroskopisch.[1][1]

Application

Guanidiniumchlorid (GdmCl) wird als Bestandteil der phosphatgepufferten Kochsalzlösung (PBS) für die Inkubation von Schlemm-Kanal-Zellen verwendet.[3] Es wird außerdem als Bestandteil des Aufschlusspuffers für die Gelierung und Expansion humaner HeLa229-Zellen verwendet.[4] GdmCl wird auch zur Isolierung von RNA eingesetzt.[5]

Stark chaotropes Mittel zur Denaturierung und nachfolgenden Neufaltung von Proteinen. Dieses starke Denaturierungsmittel kann unlösliche oder denaturierte Proteine, wie z. B. Einschlusskörper, lösen. Dies kann als erster Schritt bei der Neufaltung von Proteinen oder Enzymen in ihre aktive Form eingesetzt werden. Harnstoff und Dithiothreitol (DTT) sind ggf. auch erforderlich.

Biochem/physiol Actions

Guanidiniumchlorid (GdmCl) ist bei Konformationsänderungen und Verlust der Enyzmaktivität aufgrund einer Inaktivierung des Enzyms beteiligt.[2] Es ist am Aktivitätsverlust des Enzyms zur alkalischen Phosphatase (ALPase) bei Haliotis diversicolor beteiligt.[2] GdmCl funktioniert durch die Behinderung der Aktivität der Adenosintriphosphatase (ATPase) des Hitzeschockproteins 104 (Hsp104) in vivo.[6] Es kann außerdem Enzyme wie Papain und Aminocyclase inaktivieren.[1] Höhere Konzentrationen von GdmCl führen zur viralen Inaktivierung des Herpes-simplex-Virus 1 (HSV-1).[7]
