General description

5-(und 6-)Carboxyfluoresceindiacetat-N-Succinimidylester (CFSE) ist ein fluoreszierender Farbstoff, der membrandurchlässig ist. Er markiert Zellen, indem er kovalent an intrazelluläre Moleküle bindet. CFSE wird zur Verfolgung der Migration und Proliferation von Lymphozyten eingesetzt.

Application

5-(6-)Carboxyfluoresceindiacetat-N-Succinimidylester (CFSE) ist eine Amin-reaktive fluoreszierende Sonde, die zur Färbung von Zellen, insbesondere Immunzellen, für Analysen in Anwendungen wie der Durchflusszytometrie verwendet wird.[1][2] Bildet bei Deacetylierung grün fluoreszierende Konjugate.
Amin-reaktives Label für die Langzeit-Überwachung von Zellen. Bildet nach Deacetylierung grün fluoreszierende Konjugate.
Verwendung von 5-Carboxyfluoresceindiacetat-N-Succinimidylester:
  • zur Markierung von Zellen für die Durchflusszytometrie
  • bei Proliferationsassays von Lymphozyten
  • zur Bestimmung des intrazellulären oder intraorganellen pH-Werts in Chloroplasten
  • zur Markierung von Leishmania amazonensis und Leishmania major

Biochem/physiol Actions

5-(6-)Carboxyfluoresceindiacetat-N-Succinimidylester wird als Indikator für die Esterase-Aktivität verwendet.

  1. Hi. I bought your CFSE. I want to ask how to use it? I follow one paper: doi: 10.1016/j.xcrm.2025.102027. They used CFSE in the final concentration of 0.5 uM for 3 mins at RT. Is this you suggested? And if yes, how to prepare 0.5 uM?

    1. No direct reconstitution protocol is available for this product. Based on an online literature search, the CFSE dye, purchased as carboxyfluorescein diacetate succinimidyl ester (MW 557.47 g/mole), is typically provided in 25 mg vials. To prepare a final stock solution of 5 mM, dissolve the 25 mg in 8.96 mL DMSO. This solution can be stored as 50-100 μL aliquots at -20 °C for several months. It is important to avoid exposure to aqueous solutions during storage. Note that the CFSE dye is at a final concentration of 5 μM, however, the optimum concentration may have to be determined for each batch prepared, and then checked every 6 months to ensure it is effective.

    1. No. This compound and conjugates of this compound become fluorescent only when acted upon by an enzyme such as esterase. The enzyme removes the acetate groups.
