Beschreibung

General description

Blut-Hirn-Schranken-Permeabilisator, eine nicht toxische, tertiäre Amidverbindung, die als hoch-affiner, potenter, multimodaler Blocker der RAGE (Rezeptor für Advanced Glycation Endproducts) V domänenvermittelten Ligandenbindung wirkt (K_i = 25, 148 und 230 nM jeweils gegen Aβ;40, HMGB1 und S100B, an sRAGE bindend). Blockiert den RAGE-vermittelten Influx von Aβ;40 und Aβ;42 in das Gehirn. Unterdrückt auch die Aβ;-RAGE-induzierte NF-κ;B-Aktivierung und die NF-κ;B-abhängige Transkription der β;-Sekretase. Berichten zufolge hat die tägliche Behandlung eines murinen APP^sw/0-AK-Modells (1 mg/kg/Tag intraperitoneal) eine wesentliche Reduzierung von Thioflavin-S-positiven Amyloid-Plaques im Kortex und Hippocampus (um 70 bis 80 %) bewirkt und die kognitive Performance auf die Ebene von nicht von Alzheimer-Krankheit befallenen Mäusen gebracht.
