Beschreibung

General description

Ein zellpermeables Oxodihydrochinolin, das bevorzugt die Transkriptionsaktivität des Forkhead Box O-Familienmitglieds Foxo1 gegenüber den funktionell verwandten Foxo3a und Foxo4 hemmt (70 %, 20 % und 3 % Hemmung gemäß HepG2-basierten Reporter-Assays; [AS184256] = 100 nM); dies erfolgt dosisabhängig (IC₅₀ = 33 nM) mittels direkter Bindung des Nicht-Ser256-phosphorylierten/aktiven Foxo1, jedoch nicht der Ser256-phosphorylierten/inaktiven Form von Foxo1. Hemmt nachweislich die Gluconeogenese in Ratten-Hepatom-Fao-Kulturen (IC₅₀ = 37 und 130 nM gegen PEPCK und G6Pase-mRNA-Wert; IC₅₀ = 43 nM gegen Glucoseproduktion) in vitro und in Mäuseleber sowohl von normoglykämischen ICR-Mäusen (60 % bzw. 45 % Hemmung der hepatischen G6Pase und PEPCK-mRNA-Werte; drei 100 mg/kg orale Dosen in Stunde 0, 12, 24) als auch diabetischen db/db-Mäusen (86 %, 70 %, 56 %, 48 %, 35 % und 31 % Hemmung hepatischer G6Pase bzw. ABCG8, ABCG5, PEPCK, IL-1β; und apoCIII-mRNA-Werte; drei 100 mg/kg orale Dosen in Stunde 0, 12, 24) während einer 26-stündigen Fastenzeit in vivo.
