Specificity

Clone 10KS is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects Breast cancer resistance protein (BCRP). It targets an epitope within the N-terminal cytoplasmic domain.

Immunogen

His-tagged recombinant fragment corresponding to 126 amino acids from the N-terminal, cytoplasmic domain of human Breast cancer resistance protein (BCRP).

Application

Anti-BCRP (ABCG2), clone 10K5 ZooMAb, Cat. No. ZRB1217, is a highly specific rabbit recombinant monoclonal antibody that targets BCRP (ABCG2) and has been tested for use in Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected BCRP (ABCG2) in human placenta and mouse placenta tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected BCRP (ABCG2) in HepG2, A549 and JEG-3 cells.

Note: Actual optimal working dilutions must be determined by end user as specimens and experimental conditions may vary.

Target description

ATP-binding cassette sub-family G member 2 (UniProt: Q9UNQ0; also known as Breast cancer resistance protein, BCRP, CDw338, Mitoxantrone resistance-associated protein, Placenta-specific ATP-binding cassette transporter, Urate exporter, CD338) is encoded by the ABCG2 (also known as ABCP, BCRP, BCRP1, MXR) gene (Gene ID: 9429) in human. BCRP is a multi-pass membrane protein of the ABC transporter superfamily. It consists of six transmembrane domains and can functions as a homodimer or homotetramer. It contains an ABC transporter domain (aa 37-286) and an ABC transmembrane type-2 domain (aa 389-652). Unlike most of the other ABC transporters, BCRP contains only one nucleotide binding domain (aa 80-87). BCRP is highly expressed in placenta and lower expression is reported in small intestine, liver, and colon. It is also expressed in stem cells of different tissues and may be used as stem cell marker. It serves as a high-capacity urate exporter that functions in both renal and extrarenal urate excretion. It also serves as a xenobiotic transporter that plays an important role in the exclusion of xenobiotics from the brain. BCRP is overexpressed in many hematological malignancies and in solid tumors and contributes to resistance to a variety of anticancer agents. It has been implicated in the efflux of numerous drugs, including camptothecin, methotrexate, azidothymidine (AZT), daunorubicin, and doxorubicin. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
