General description

AF4/FMR2 family member 3 (AFF3), a tissue-restricted nuclear transcriptional activator, is mainly expressed in the lymphoid tissue. This RNA binding protein is a member of the AF4/LAF4/FMR2/AF5q31 family. AFF3 is a component of the transcriptional super elongation complex. It is expressed at a high level in the thymus. AFF3 has two domains with transactivation activity. This lymphoid-specific gene, is located on the human chromosome 2q11.2.

Biochem/physiol Actions

AF4/FMR2 family member 3 (AFF3) participates in transcriptional regulation. It is a potential clinical modulator of estrogen receptor (ER) signaling. This transcription activator can play a role in lymphoid development and cancer. It can bind to double-stranded DNA in vitro. AFF3 overexpression causes tamoxifen resistance and estrogen-independent proliferation in ER+ human breast tumors.
