General description

Hemin is a pro-oxidant, hemolysis derived-product. It is essential for the growth of heme-deficient organisms such as trypanosomatids. Hemin readymade solution is used as a culture-medium supplement for fastidious anaerobic microorganisms, shown to enhance the growth of many anaerobes including Bacteroides species and certain gram-positive non-spore forming organisms. It is present as a key ingredient compound in common anaerobic growth media, such as Brucella broth, Thioglycolate broth and Reinforced Clostridial Medium.

Application

Hemin solution may be used as a supplement in brain heart infusion-yeast extract-hemin (BHI-YH) medium to determine cholesterol-to-coprostanol reducing activity.

Other Notes

  1. For 100 ml of medium preparation, it is recommended to add 1 mL hemin ready-made solution after autoclaving the medium, to achieve a final concentration of 5 microgram/ml. The optimal concentration of the supplement may vary for each strain.
  2. The solution is classified as light sensitive. 2. The solution should not be heated and must be added aseptically to autoclaved media cooled to 50-55°C. 3.
  3. Select strains which show turbid growth (24-48 hr.) with Hemin supplementation: Porphyromonas gingivalis, Bifidobacterium longum, Clostridium perfringens, Bacteroides vulgatus and Fusobacterium nucleatum.
