Application

Human AB serum is used in tissue engineering, transplantation and cell therapy applications for the expansion of mesenchymal stem cells (MSC) from adipose tissue or mesenchymal stromal cells from human bone marrow; for standardized limbal epithelial stem cell graft generation and transplantation; for ex vivo expansion of NK cells from peripheral blood in Hematopoeitic Stem Cell Expansion Medium; and for upgrading pretransplant human islet culture technology.
Human Serum has been used: as a component of phosphate-buffered saline with Tween® detergent (PBST) in bridging enzyme-linked immunosorbent assay (ELISA), as a mock biological sample for assessing the performance of paper-based ELISA assay with complex biological samples, as a diluent to create a dilution series of the baseline sample for the generation of standard curve for NISTmAb spiked into serum, as well as for multiple myeloma (MM) patient sample MM2232

Biochem/physiol Actions

Human AB serum is typically obtained by allowing whole blood to clot naturally, which is known as Off-The-Clot (OTC) serum. Human serum obtained from clotted whole blood is preferred as it usually contains higher levels and a more diverse range of growth factors for optimal cell growth. Human serum from blood-type AB donors lacks antibodies specific for both A and B blood-type antigens. This makes it a universal donor serum that is suitable for various cell culture types including human stem cell lines.

Packaging

Packaging may vary for different batches due to short supply of the bottles used to pack this product. The presence or lack of shrink wrap around the cap of the bottle doesn’t impact product integrity/sterility of the product.

Analysis Note

Endotoxin tested

Other Notes

It is not unusual for our human serum products to be cloudy. This product is from pooled serum and cloudiness indicates that lipids are present in the serum. No clear, lipid-free human serum products are available.

Disclaimer

Each donor is tested for and found non-reactive for Hepatitis B & C and non-reactive for Human Immunodeficiency Virus (HIV) antibody by ELISA. Nevertheless, products of human origin should be considered potentially infectious and handled accordingly.

Questions

1–10 of 29 Questions
  1. In dem Text der Produktbeschreibung steht, dass das Serum durch off-the clot Gerinnung und Zentrifugation gewonnen wird. In dem Kommentar von Fred steht, dass es aus citrate plasma durch zufügen von calcium gewonnen wird. Was davon stimmt?

    1 answer
    1. The initial starting material used to prepare this product is citrated to prevent clotting. The whole blood is centrifuged and the plasma is separated. Calcium is then added to the plasma fraction to initiate clot formation. The resulting supernatant is then centrifuged to remove cellular debris.

  2. Is there a significant difference in composition between different source batches (SLCQ2795 and 0000523739) of the same H4522-100ML serum? I used it to simulate human serum for detecting the EC50 values of compounds.

    1 answer
    1. This serum product is pooled from multiple donors. Typical pooled batches range from 50 – 70 donors, but large batches may have up to 200. While some degree of variability from lot to lot exists, each lot is required to meet published test specifications. Please refer to the Certificate of Analysis to compare lot-specific results, and the Specification Sheet to review product specifications:

  3. Does this product contain Estradiol or progesterone or is is already stripped

    1 answer
    1. This product is not charcoal-stripped or hormone-depleted. As a human serum product derived from pooled male plasma from blood group AB, this product may contain naturally occurring levels of estradiol and progesterone. Concentrations of estradiol and progesterone are not listed as tested specifications and are therefore not provided.

      Helpful?

  4. Is this product obtained from healthy individuals? I noted that each donor unit used in the preparation was tested and found negative for HBsAG, HIV and etc. However, does each donor was tested for any underlying diseases like Diabetes and medically fit?

    1 answer
    1. This serum product is pooled from multiple donors. Typical pooled batches range from 50 – 70 donors, but large batches may have up to 200. The general health and/or underlying condition information is not collected from individual donors. Based on electrophoresis studies, the pooled final product may be considered ‘Normal’ serum.

  5. 안녕하세요, 혹시 해당 제품의 -20도에서 보관기간을 알 수 있을요?

    1 answer
    1. This product should be stored at -20°C. The storage period at this temperature is valid up to the lot-specific expiration date printed on the product label or COA. The information requested is lot-specific and reported in the Certificate of Analysis. Please see the link below to access a Certificate of lot interest:

  6. Is this serum contain Uric acid? If yes how much, or how to check it?. If the serum is expired, is it okay to use?

    1 answer
    1. While uric acid is present in human serum, this product is not tested for uric acid levels. Should uric acid measurement is required, see product MAK483, Uric Acid Assay Kit, which is suitable for the determination of Uric acid in serum. It is not recommended to use products that are beyond the assigned expiration date.

  7. Once the product has been thawed, how should it be stored? How many times can be freezed and thawed?

    1 answer
    1. After the initial thaw, the serum can be aliquoted in to single use vials and stored at -20°C. For more information, please refer to the following page:
