Gene Information

human ... PDPK1(5170)

General description

FUNCTION: SwissProt: O15530 # Phosphorylates and activates not only PKB/AKT, but also PKA, PKC-zeta, RPS6KA1 and RPS6KB1. May play a general role in signaling processes and in development (By similarity). Isoform 3 is catalytically inactive.
SIZE: 556 amino acids; 63152 Da
SUBCELLULAR LOCATION: Cytoplasm. Membrane; Peripheral membrane protein. Note=Membrane-associated after cell stimulation leading to its translocation. Tyrosine phosphorylation seems to occur only at the plasma membrane.
TISSUE SPECIFICITY: Appears to be expressed ubiquitously.
PTM: Phosphorylated on tyrosine and serine/threonine. Phosphorylation on Ser-241 in the activation loop is required for full activity. PDK1 itself can autophosphorylate Ser-241, leading to its own activation.
SIMILARITY: SwissProt: O15530 ## Belongs to the protein kinase superfamily. AGC Ser/Thr protein kinase family. PDK1 subfamily. & Contains 1 PH domain. & Contains 1 protein kinase domain.

Quality

Evaluated by Kinase assay: Enzyme reaction is conducted in a buffer containing 50 mM HEPES (pH7.5), 10 mM MgCl2, 1 mM EGTA, 200 EM ATP, 0.01% Brij-35 and 2 EM substrate (SER/THR 6, Invitrogen) at room temperature for 1 hour.

Physical properties

89 kDa

Physical form

40 mM Tris-HCl, pH 8.0, 110 mM NaCl, 2.2 mM KCl, 20% glycerol, and 3 mM DTT

Storage and Stability

Stable for > 6 month at –80°C from date of receipt.
