Specificity

Clone 1C4 is a ZooMAb® rabbit recombinant monoclonal antibody that detects DLX2. It targets an epitope within 16 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 16 amino acids from the N-terminal half of human DLX2.

Application

Quality Control Testing

Evaluated by Western Blotting in PC-12 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected DLX2 in PC-12 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected DLX2 in mouse brain lysates and DLX2 recombinant protein.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected DLX2 in HEK-293 cells.

Affinity Binding Assay: A representative lot of this antibody bound DLX2 peptide with at least ten thousand-fold (10,000X) higher than with non-specific control peptide.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected DLX2 in human prostate tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Homeobox protein DLX-2 (UniProt: Q07687; also known as DLX-2, also known as distal-less homeobox 2) is encoded by the DLX2 gene (Gene ID: 1746) in human. DLX2 is part of the DLX family of transcription factors that share homology with the Drosophila distal-less (Dll) gene. DLX-2 is characterized by its homeodomain, a DNA-binding domain consisting of 60 amino acids that facilitate the binding of DLX-2 to specific DNA sequences to regulate gene expression. It may also contain other domains or motifs that influence its interaction with co-factors and other components of the transcriptional machinery. DLX-2 is involved in the regulation of embryonic development, particularly in the formation of the forebrain, facial structures, and limbs. It acts as a transcription factor that can activate or repress the expression of genes, playing a crucial role in morphogenesis and differentiation. The expression of DLX-2 is primarily observed during embryonic development. It is expressed in specific patterns, particularly within the developing forebrain and branchial arches, regions that give rise to facial structures. After development, DLX-2 expression is significantly reduced and is present at low levels in adult tissues. DLX-2 has been associated craniofacial abnormalities, such as cleft palate and other craniosynostosis syndromes, Neurodevelopmental Disorders like autism spectrum disorder (ASD) and intellectual disabilities. There is evidence suggesting that DLX-2 may be involved in tumorigenesis. It has been reported that DLX-2 can promote the proliferation of osteosarcoma cells and may be involved in the progression of other types of cancer.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
