Specificity

Clone 4D17 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Nanog. It targets an epitope within 20 amino acids from the N-terminal half. Predicted to react with both isoforms.

Immunogen

KLH-conjugated linear peptide corresponding to 20 amino acids from the N-terminal half of mouse Nanog.

Application

Quality Control Testing

Evaluated by Western Blotting in E14-TG2a cell lysates.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Nanog in E14-TG2a cell lysates.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Nanog in Mouse embryonic stem cell lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Nanog in mouse testis tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Nanog in E14-TG2a cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Homeobox protein NANOG (UniProt: Q80Z64; also known as ES cell-associated protein 4, Early embryo specific expression NK-type homeobox protein, Homeobox transcription factor Nanog) is encoded by the Nanog (also known as Ecat4, Enk) gene (Gene ID: 71950) in murine species. Nanog is a transcription factor that is involved in the self-renewal of embryonic stem cells (ES) and is a critical factor for the maintenance of the undifferentiated state of pluripotent cells. It binds optimally to the DNA consensus sequence 5′-TAAT[GT][GT]-3′ or 5′-[CG][GA][CG]C[GC]ATTAN[GC]-3′. Its expression is observed in inner cell mass (ICM) of the blastocyst and gonocytes and its expression decreases with ES differentiation. Nanog can autorepress its own expression in differentiating ES cells by binding to its own promoter following interaction with ZNF281/ZFP281, leading to recruitment of the NuRD complex and subsequent repression of expression. Overexpression of Nanog in ES cells maintains specific differentiation, however, its overexpression in cancer stem cells is reported to inhibit apoptosis. Together with OCT4, it can activate SNAIL2 and promote metastasis and the EMT phenotype in cancer cells. Nanog undergoes phosphorylation on serine 130 and 131 by casein kinase I and phosphomimetic substitution of serine 130 is shown to abolish its DNA binding and eliminates LIF‐independent self‐renewal. On the other hand, phosphomimetic substitution of serine 131 enhances LIF‐independent self‐renewal but does not affect its DNA binding. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
