Información sobre el gen

Descripción general

Our Alpha 1 Antitrypsin is a human plasma protein. It is shown to be HBsAg, HCV, HBc free and also negative for anti HIV1 and HIV2 according to FDA approved tests.
The gene AAT (α 1 antitrypsin) is mapped to human chromosome 14q32.1.[1] AAT is an acute-phase plasma protein.[2] It is a water soluble glycoprotein. AAT is mainly secreted by hepatocytes, macrophages, and some cancer cells.[3] AAT is also referred to as Serpin A1.

Aplicación

AAT (α 1 antitrypsin) from human plasma has been used as a standard to check the presence of AAT in benign and malignant lymphoreticular cells and tissues.[4]

Acciones bioquímicas o fisiológicas

AAT (α 1 antitrypsin) functions as a serine protease inhibitor with targets like elastase, plasmin, collagenase, thrombin, leucocyte proteases, trypsin, chymotrypsin and plasminogen activator.[3][5] Clinically, its deficiency is associated with two major diseases: pulmonary emphysema and early onset/juvenile hepatic cirrhosis.[3][6] It is elevated in inflammatory conditions, malignancies, liver disease and pregnancy and also after surgical trauma and use of oral contraceptives.[7][8] The simultaneous quantitative determination of AAT and ceruloplasmin permits differential diagnosis of liver afflictions. Rise in AAT occurs as an acute phase response to tissue necrosis and inflammation.[9] It might work as a biomarker for cutaneous squamous cell carcinoma, hepatitis B, insulinomas, NSCLC (non-small cell lung cancer), papillary thyroid carcinoma, lung cancer and breast carcinoma.[10]

Questions

1–2 of 2 Questions
  1. How is the amount of Alpha 2-Antiplasmin in the vial supplied (lyophilized) determined? Specifically, which assay is used to determine the amount of Alpha 2-Antiplasmin (100 ug)?

    1 answer
    1. The protein concentration is measured by its absorbance at A280, utilizing an extinction coefficient of 0.703 for a 0.1% solution at a 1 cm path length.

      Helpful?

  2. What is the correct molecular weight for alpha 2 antiplasmin?

    1 answer
    1. The predicted molecular weight of human alpha-2 antiplasmin, based on its amino acid sequence, is approximately 50,500. However, when 11-14% carbohydrate content is considered, the molecular weight increases to a maximum of about 58,000. Despite this, physical measurements of the molecular weight have varied from 63,000 to 70,500.
