General description

Polo-like kinase 1 (PLK1) belongs to the polo-like kinase family. PLK1 is characterized with two COOH-terminal polo box domain and N-terminal kinase domain involved in drug development. PLK1 is expressed primarily at centrosomes and at the midbody of dividing cells.

Immunogen

The antiserum was produced against synthesized peptide derived from human PLK1 around the phosphorylation site of Ser137.

Immunogen Range: 103-152

Biochem/physiol Actions

Polo-like kinase 1 (PLK1) is involved in regulation of mitosis. PLK1 plays a crucial role in centrosome-mediated microtubule events and spindle assembly. PLK1 expression is elevated in cancer tissues. Thus, it can be used as a potential prognostic marker for several cancers, including non-small-cell lung cancer, squamous-cell carcinomas of the head and neck, melanomas, colorectal cancer, oropharyngeal carcinomas, and ovarian and endometrial carcinomas. PLK1 recruited to centrosomes during S-phase by centrosomal protein FOP-related protein of 20kDa (FOR20), plays a vital role in S-phase progression. The protein also plays an essential role in central spindle formation and cytokinesis. PLK1 inhibitors might act as a potential therapeutics in cancer treatment.

Centrosomes are required for efficient cell cycle progression mainly by orchestrating microtubule dynamics and facilitating G1/S and G2/M transitions. However, the role of centrosomes in S-phase progression is largely unknown. Here, we report that depletion of FOR20 (FOP-related protein of

Human polo-like kinase 1 (PLK1) is essential during mitosis and in the maintenance of genomic stability. PLK1 is overexpressed in human tumours and has prognostic potential in cancer, indicating its involvement in carcinogenesis and its potential as a therapeutic target.

The human protein kinase Plk1, a member of the polo-like kinase family, is known to function at mitosis. Here we show that the relative specific activity of Plk1 increases in mitosis, that Plk1 is specifically phosphorylated during mitosis, and that
