General description

Monoclonal Anti-phospho-RasGRP1(pThr184) (mouse IgG2a isotype) is derived from the hybridoma p-RasGRP1-1 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with a synthetic phospho-peptide corresponding to residues surrounding Thr184 of human RasGRP1 conjugated to KLH. RAS guanyl releasing protein 1 (RasGRP1) is a member of RasGRP family, characterized by the presence of a N-terminal Ras superfamily guanine nucleotide exchange factor (GEF) domain and and C-terminal calcium- and diacylglycerol (DAG)/phorbol ester-binding domains. It is an intracellular signaling protein. RasGRP1 has a N terminal guanine nucleotide exchange factor domain for Ras-like guanosine triphosphatases (GTPases) (RasGEFN/RasGEF), two EF hand motifs and a carboxy-terminal C1 domain.

Immunogen

a synthetic phospho-peptide corresponding to residues surrounding Thr184 of human RasGRP1, conjugated to KLH. The corresponding sequence is identical in rat, mouse, monkey, bovine and canine RasGRP1.

Monoclonal Anti-phospho-RasGRP1(pThr184) antibody produced in mouse has been used in immunoblotting and immunoprecipitation.

Biochem/physiol Actions

RAS guanyl releasing protein 1 (RasGRP1) functions as a diacylglycerol (DAG)-regulated nucleotide exchange factor specifically activating Ras through the exchange of bound guanosine diphosphate (GDP) for guanosine triphosphate (GTP). It activates the extracellular signal-regulated kinase (Erk)/mitogen-activated protein (MAP) kinase cascade and regulates T-cells and B-cells development, homeostasis and differentiation.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
