General description

Mast cells play an important role in innate immunity or the response of the body to invading pathogens. The functions of mast cells are regulated by a number of surface receptors; the receptors to Stem cell factor (SCF; c-Kit ligand or mast cell growth factor) being the most critical. The signalling triggered by SCF results in mast cell activation in response to antigens. The response involves tyrosine phosphorylation by specific tyrosine kinases that drive the downstream signalling process. The secondary pathways activated by SCF are signalling mediated by Akt, mTOR, MAPK and c-Jun. In addition to development of mast cells, SCF also regulates their subsequent maturation and homing to the residence sites. Constitutive activation by SCF is implicated in myeloproliferative discorders, systemic mastocytosis and gastrointestinal stomal cell tumors
Anti-Stem Cell Factor antibody reacts specifically with mouse stem cell factor.

Specificity

The antibody will neutralize the biological activity of recombinant mouse SCF.

Application

Anti-Stem Cell Factor antibody may be used to neutralize SCF at a concentration of 25 ng/ml in 1X105/ml TF-1 cells. The antibody has been used to neutralize SCF in primary murine embryonic fibroblast (MEF) culture. The recommended concentration for indirect ELISA is 1.0 μg/ml and for immunoblotting is 2.0 μg/ml.
