General description

The p190 RhoGAP protein is also termed as p190A or glucocorticoid receptor binding factor 1 (GRLF1) 190 kDa. It has three major regions, such as an N-terminal GTP binding domain, a middle region and a C-terminal GTPase-activating proteins (GAP) domain. Rho GTPase activating protein 35 (ARHGAP35) codes for p190A RhoGAP (p190A) protein. It is an important GTPase-activating protein (GAP), which belongs to the Rho GTPase-activating proteins (RhoGAP) family. This gene is located on human chromosome 19q13.3

Specificity

Anti-p190 RhoGAP (GK-18) specifically recognizes p190 RhoGAP (190 kDa).

Immunogen

synthetic peptide corresponding to amino acids 1131-1148 located near the C-terminus of rat p190 RhoGAP, conjugated to KLH.

Application

Anti-p190 RhoGAP (GK-18) antibody produced in rabbit may be used in immunoblotting and immunoprecipitation.

Biochem/physiol Actions

The p190 RhoGAP protein or glucorticoid receptor binding factor 1 (GRLF1) serves as a GTPase-activating protein (GAP) for the Rho and Rac family of proteins, that are involved in regulating the reorganization of cytoskeletal actin and membrane ruffling. It controls axonal growth and guidance. p190 RhoGAP is essential for normal neural development. p190 RhoGAP has been shown to be tyrosine phosphorylated by c-Src and v-Src both in vitro and in vivo. The middle domain region of this protein mediates binding to Ras GTPase activating protein (p120 RasGAP). p190A plays a key role in the development of mouse and tissue homeostasis. In mammals, p190A also participates in the proliferative and motile capacities of cells. Mutations in the gene is implicated in cancer.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 1% BSA and 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8 °C for up to one month. For extended storage freeze in working aliquots. Repeated freezing and thawing is not recommended. Storage in "frost-free" freezers is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
