Descripción general

Kelch-like protein 12 is a novel human kelch protein localized to a ring canal structure. It consists of N-terminal POZ (poxviruses and zinc fingers) and intervening region (IVR) domains. It plays an important role in the ring-like structure activity during oocyte development.

Acciones bioquímicas o fisiológicas

Kelch-like protein 12 is a protein encoded by the KLHL12 gene in humans. It is a Bric à Brac (BTB)-Kelch protein and specifically binds to the polymorphic region of the D4 receptor through its Kelch domain. KLHL12 also interacts with Cullin3 and functions as an adaptor to target the D4 receptor to an E3 ubiquitin ligase complex. Its overexpression strongly promotes ubiquitination of the D4 receptor. KLHL12 is essential for the development of the ring-like structure, which is independent of the ability to bind actin. It interacts with both immature, ER-associated and mature, plasma membrane-associated D4 receptors, β-arrestin2, and also promotes ubiquitination of both receptor subpools.
